Hi, welcome to HCM,
Incubation period for genital herpes is 4-5 days (approx.), and most common site is foreskin, glans and shaft of the penis (not scrotum). Further herpes classically presents as small fluid filled blister which appears in group and common symptom is burning.
As per your description you have a red spot on scrotum which are itchy. Therefore to my knowledge, this is unlikely to be genital herpes. I would consider scrotal dermatitis more likely. However ELISA based serological tests is the only means to confirm or rule out herpes infection. You may consider this test.
