As I said earlier masturbation is completely harmless and do not affect in any way including sperm count. Complaint of low back pain and knee pain are indicative of low hemoglobin or other pathology rather than masturbation. There is also possibility of some joint problem so checking with rheumatologist is highly suggested.
Before you want to increase the sperm count you need to know what is current level.
For dong same you can get a lab report of semen analysis from nearby laboratory. It is simple and cheap test and reveals number of sperms in semen.
Regarding your comment about prescription I must make it clear that they are just for information and not a prescription. The normal time to discharge is subjective thing and there is no time limit. The discharge which leaves you and your partner unsatisfied is called premature ejaculation. In most persons this time is less than two minutes and any discharge occurring before two minutes is called premature.
