Question : I have a lump behind my right knee. It is somewhat painful when I flex it. My concern is that I sit all day and it might be a blood clot.
I just came back from a long walk and noticed it about half an a hour after I sat down at my desk.

It could be a Bursa

As per your description, it could be a Bursa.
There will be Communication to the knee joint.
It becomes painful if it is infected.
Spontaneous collection of Blood leading to Haematoma is rare.It can happen in Coagulation disorders.
Ultrasound/Colour Doppler of the area will give a Clue.
Please consult a Surgeon/Orthopedician and proceed further.
