Question: I was recently prescribed one dose of Azithromycin for mycoplasma Genitalium, how long should I wait after taking the medication before getting tested again

Genital Mycoplasma infection is usually diagnosed by ruling out the common causes of genital infections like Gonorrhea, Chlamydia, and Trichomonas.
Test for cure usually not required. Taking the drugs for 2 weeks and the relief of symptoms will assure that you are free from Mycoplasma genitalium infection. Urine deposits examined for the presence of pus cells will help in most cases. If it remains less than 4 pus cells, it assures that you had been cured.
Moreover it depends upon what type of test was used to diagnose the Mycoplasma infection. If it is through isolation of organism, then repeat culture will show no growth after the treatment.
If it is through serological test then depend upon whether it is a antibody detection test (IgG) it will be persisting even after treatment.
If it is through PCR or NAAT it will take not less than few months to show a negative test.

The test was done with a PCR test. So even after treatment and relief of symptoms I could still test positive on a PCR test for a few month? Does that mean I could still infect others even if the infection symptoms have cleared

I had much experience with HIV PCR results and follow up. The course of HIV is entirely different from that of Mycoplasma Genitalium which is a bacterial diseases and amenable to antibiotics. I don't have any experience with mycoplasma PCR.
So it will be better for you to clarify the same from a microbiologist about when to repeat the test after the treatment in case of Mycoplasma Genitalium infection.
