Yes, just a single injection of benzathine penicillin 2.4 MU, will cure syphilis. This is recommended for treating primary syphilis. However, if the individual is allergic to penicillin or for some other reasons, there are alternative treatments:
Alternative treatment options are:
-Azithromycin as a single 2 gm oral dose is effective for treating Primary syphilis.
-Doxycycline 100 mg orally twice daily for 14 days is also effective.

Do I have to apply both together ( Azithromycin and Doxycycline ) ? or one of them enough ?
If the option Azithromycin, Do I need to repete the dose after 3 days for example for guarantee the result 100 % ?

Just, one of them is enough. i.e either Azithromycin 2 gm, single oral dose OR Doxycycline 100mg twice daily, orally, for 14 days.
No need to repeat Azithromycin.
However, you must get a prescription for these from a doctor, because they are prescription medicines.

No doubt, this report mentions a few treatment failures, however this is not widespread, and moreover CDC guidelines still says that a 2-gram dose of azithromycin may be considered for penicillin-allergic patients. However, single injection of benzathine penicillin 2.4 MU, is the preferred and recommended treatment by CDC as well as WHO, for treating primary syphilis (as i told you before).

Hi.
Tetracycline is also an alternative treatment for early stage syphilis, dose is 500mg, every 6 hours for 2 weeks. It can also be given in penicillin allergic patients just as doxycycline or azithromycin. However because it requires more frequent dosing(compliance issues) and also produces GI side effects, therefore doxycycline is preferred over it.
