Brief Answer:
Get your wife further tests as following

Detailed Answer:
Hi,

This is Dr.Sejdini. I have just received your query and I'm here to help.

Having itchiness when having sex with your wife, which is not present after wearing condom, is indicating that your wife might be having vaginal problems (possible fungal infection).

That is especially true if she is having vaginal discharge too? itching? burning? pain especially when having sex?

If I was your caring doctor would advise you to run further tests like vaginal swab test, urine analysis.

Treatment depends on the tests results, locally (per vaginal) and orally.

Hope this helps.
Let me know if you have further questions.
