Gemcitabine replaces cytidine, during DNA replication.
Hence the DNA cannot produce the required protein and the cell dies

It also binds to ribonucleotide reductase (RNR) active site and inactivates the enzyme irreversibly. Hence deoxyribonucleotides required for DNA replication and repair are not produced
