Your husband will need the following tests to be done :
1. Get examined by an Urologist, who'll perform an examination, including DRE.
2. urine routine and culture test.
3. blood routine, creatinine, sugar, LFT, platelets, BT,CT and PT.
4. ultrasound of abdomen-KUB, with TRUS( trans rectal ultrasound scan ).
After seeing theese reports, a good idea of the diagnosis can be made out.
If a malignant nodule is felt on DRE, and/or a suspicious lesion on TRUS of prostate is seen, ultrasound guided needle biopsy, will need to be done.
The biopsy will confirm the diagnosis.

Yes, there're several causes for PSA to go up,like :
1.Prostatitis
2.Large prostate gland
3.Following prostate gland manipulation, such as instrumentation or digital massage.
4. cancer prostate.
This is the reason, why an Urologist should do the DRE examination and
