albumin is a fraction of protein (it is one type of protein). normally in kidney diseases protein leaks in to urine . it can be albumin or any other protein. if we do only albumin testing we can miss some other protein which may be there in urine. so protein/creatinine ratio is a better option.
most labs now use modified jaffe kinetic method for testing, in any case any reading above 1.2 should be considered as abnormal and all efforts should be made to identify the problem. kidney disease can occur in normal range of creatinine also.

Generally we don't collect a 24 hrs urine sample for protein creatinine ratio. we mostly do spot urine - that means a one time urine sample of around 10 ml.
but if you want to collect a 24 hrs sample. then you have to collect whatever you pass throughout the day whether it is 2 litre or 5 litre into the container. creatinine can be decreased by precautions as explained. if it decreases it means kidney is working better
